Still, I prefer the idea of the game being single-player, perhaps we will finally have the 40k-non Spess Mehreen RPG we (or maybe just me) always wanted?

There hasn't been Necrons in any W40k game since DC, and that was more than 5 years ago now.

It would be surprising to discard the work Vigil have probably done on the Guard/Imperial forces. They have shown a lot of screens of them thus far, but it remains to be seen if the new game will follow the same class based mechanic.
You'll be extremely lucky to get anything of living metal in this game mate. Despite the success of the latest 'Cron releases (and weirdo fluff), I don't think that THQ see them as a suitable faction to use in game, not sure why?

Indeed, I would be surprised to see no IG either, but they're not 100% definite like the others.
GW, and conversely THQ and Relic are both extremely biased towards Space Marines, purely because that's what the majority audience is. It makes sense why it is the case, but doesn't make it any less annoying considering 95% of releases are focussed on the same race. The thing that gives Warhammer and W40k depth is the incredibly variety and differences between the races. If I wanted to watch absurdly masculine men in power armour murdering other absurdly masculine men in power armour I'd watch some generic scifi.
Regardless, I am quietly optimistic. Hopefully someone non-Relic will be a breath of fresh air, if it ever sees the light of day.

How would that work in a 40k singleplayer RPG? I mean, they all hate eachother, which would make questing impossible unless they either introduced some new "neutral" race or had multiple storylines. Which would be a huge amount of work.

At least, that's what I hope for. I don't really think THQ/Vigil want to create a Space Marine clone.
Worse case scenario I'm sure they can invent some random story like they did for the Dawn of War games, there's artefact X on this world, and all the races just happen to be on the same planet to fight over it or something.
Let's put it this way, in it's MMO state it was almost certainly going to have multiple playable races. I don't see why they'd drop such a crucial feature.
